FGF2 is a member of the Fibroblast Growth Factor family of proteins. It is often called FGF-basic or bFGF. FGF2 is a single-chain polypeptide growth factor that plays a significant role in wound healing, it is a potent inducer of angiogenesis and it is essential for the growth of many stem and progenitor cells. FGF2 shares a 55% amino acid residue identity with FGF1 and interacts with heparin sulfate with nanomolar affinity. It is a potent inducer of DNA synthesis in a variety of cell types. FGF2 activates mesenchymal splice variants of FGF receptors (c splice variants) and also activates FGF Receptor 1b, but has little activity against FGFR2b or FGFR3b. It is most closely associated with FGF1 (aFGF) (see additional information).

CCL17 is chemotactic for T-cells and plays a role in T-cell development, trafficking, and activation. It promotes atherosclerosis by regulating homeostasis of regulatory T-cells, thereby limiting their expansion. CCL17 is a ligand for CCR4, CCR8, and the erythrocyte receptor ACKR1.

CCL22 is chemotactic for chronically activated T-cells and plays a role in trafficking activated T cells. It also plays a role in migration and survival of cells expressing FOXP3. It is important for regulating IgA ASC accumulation in mammary glands and consequently the transfer of IgA antibodies from mother to infant. Expression of CCL4 is induced by IL2 and mitogens, such as phytohemmagglutinin, LPS, and the peptidoglycan wall of S. aureus and expression is inhibited by glucocorticoids, TGF-beta, and cyclosporin A. CCL4 is an HIV-suppressive factor and may contribute to amyloid deposition in Alzheimer disease. It also binds to HSV-1 virions and generates pores in the envelope of HSV-1. CCL4 is a ligand for CCR1, CCR3, CCR5, CCR8, and ACKR2 (D6).

It is expressed primarily in microglia and other macrophages, as well as myelopoietic bone marrow cells. Upregulation of expression of CCL6 has been observed after stimulation with IFNG, CSF1, CSF2, IL3, and IL4. CCL6 expression is also stimulated by IL-13 and is required for IL-13 stimulation of other chemokines. CCL6 is chemotactic for macrophages and lymphocytes and acts as a ligand primarily for CCR1. CCL6 has only been observed in rodents thus far; rat CCL6 is most homologous to human CCL14, CCL15, and CCL23.

It was first isolated from murine mast cells stimulated by IgE interacting with an antigen and is expressed and secreted by T cells, monocytes, macrophages, and a variety of tumor cell lines. Expression is induced by IFNG and inhibited by IL13. It is chemotactic for monocytes, macrophages, eosinophils, and NK cells and regulates protease secretion by macrophages which may contribute to metastasis of cancer cells. CCL7 also promotes neuritogenesis in midbrain dopamine neuron cultures. CCL7 is a ligand primarily for CCR2, as well as CCR1, CCR3, CCR10, and ACKR2 (D6).

CDNF is a secreted neurotrophic factor that contains 8 conserved cysteine residues, much like its relative MANF. CDNF and its structural homolog MANF each contain an N-terminal saposin-like lipid binding domain and a carboxyl-terminal domain, which is not homologous to previously characterized protein structures. CDNF is expressed in several tissues, including the embryonic and postnatal brains of mice, as well as the heart, skeletal muscle, and testis. It has been shown to promote survival, growth and function of dopamine specific neurons. CDNF and MANF can prevent 6-OHDA induced degeneration of dopaminergic neurons by triggering survival pathways in a rat experimental model of Parkinson disease.

Description

Ciliary neurotrophic factor (CNTF) is a polypeptide hormone whose actions appear to be restricted to the nervous system where it promotes neurotransmitter synthesis and neurite outgrowth in certain neuronal populations. It was initially identified as a trophic factor for embryonic chick ciliary parasympathetic neurons in culture. The protein is also a potent survival factor for neurons and oligodendrocytes and may be relevant in reducing tissue destruction during inflammatory attacks. In addition, CNTF is useful for treatment of motor neuron disease and it could reduce food intake without causing hunger or stress. CNTF is structurally related to IL6, IL11, LIF, and OSM. All of these four helix bundle cytokines share gp130 as a signal-transducing subunit in their receptor complexes.

Description

Colony Stimulating Factor 2 (GM-CSF) was initially characterized as a growth factor that can support the in vitro colony formation of granulocyte-macrophage progenitors. It is produced by a number of different cell types, including activated T cells, B cells, macrophages, mast cells, endothelial cells and fibroblasts, in response to cytokine or immune and inflammatory stimuli. On mature hematopoietic cells, CSF2 is a survival factor for and activates the effector functions of granulocytes, monocytes/macrophages and eosinophils. CSF2 promotes a Th1 biased immune response, angiogenesis, allergic inflammation and the development of autoimmunity. It shows clinical effectiveness in ameliorating chemotherapy-induced neutropenia, and CSF2 transfected tumor cells are utilized as cancer vaccines. Mature rat CSF2 shares 69% with mouse CSF2. Rat CSF2 is fully active on mouse cells, although Mouse CSF2 is only weakly active on rat cells.

Description

Colony Stimulating Factor 2 (CSF2), previously called Granulocyte-Macrophage Colony Stimulating Factor (GM-CSF), is secreted by a number of different cell types (including activated T cells, B cells, macrophages, mast cells, endothelial cells and fibroblasts) in response to cytokine or immune and inflammatory stimulation. It was initially characterized as a growth factor that can support the in vitro colony formation of granulocyte-macrophage progenitors and has functions of stimulates the growth and differentiation of hematopoietic precursor cells from various lineages. CSF2 has also been reported to have a functional role on non-hematopoietic cells and can induce human endothelial cells to migrate and proliferate. Additionally, it can stimulate the proliferation of a number of tumor cell lines, including osteogenic sarcoma, carcinoma and adenocarcinoma cell lines. It is reported that CSF2 has no biological effects across species.
