Description
T4 UvsX DNA Recombinase is a recombinase used in an isothermal amplification method where the recombinase complexes with the DNA primer. The recombinase-primer complex scans double-stranded DNA for a homologous sequence, which is followed by a strand exchange. During this process, the displaced strand is stabilized by bound gp32, and amplification can begin.
This isothermal reaction employing T4 UvsX DNA Recombinase does not require pretreatment of DNA, nor does it require any strand denaturation, making it suitable for field tests or performing tests with minimal equipment and supplies. Furthermore, isothermal amplification is an extremely sensitive technique.
T4 UvsX DNA Recombinase and other recombinases form presynaptic filaments on ssDNA that are activated to search for homology in dsDNA and to perform DNA strand exchange. UvsX recombinase is free from detectable nuclease activities. UvsX recombinase is the RecA/Rad51 ortholog of bacteriophage T4. Description
UvsY is the phage T4 recombination mediator protein, and structural and biophysical studies provide insights into its role in T4 homologous recombination. During T4 homologous recombination, the UvsX recombinase must compete with the prebound gp32 single-stranded binding protein for DNA-binding sites and T4 UvsY Protein stimulates this filament nucleation event. UvsY is a 15.8-kDa protein with properties that are consistent with its role as mediator; it stimulates the DNA-dependent ATPase activity of UvsX, lowers the critical concentration of UvsX that is required for activity, and promotes strand exchange. UvsY efficiently promotes the UvsX-catalyzed strand invasion reaction by recruiting to ssDNA–gp32 complexes, promotes the release of gp32, and favors the binding of UvsX. UvsY protein is free from detectable nuclease activities.
