Cephalosporins

Celphalosporins are a class of β-lactam antibiotics. β-lactams interfere with PBP (penicillin binding protein) activity involved in the final phase of peptidoglycan synthesis. PBPs are enzymes that catalyze a pentaglycine cross-link between alanine and lysine residues providing additional strength to the cell wall. Without a pentaglycine cross-link, the integrity of the cell wall is severely compromised, and ultimately leads to cell lysis and death. First generation cephalosporins are typically active against gram-positive bacteria with successive generations having an increasingly broadened activity against gram-negative bacteria. Cephalosporins are a type of β-lactam antibiotic consisting of a four-membered β-lactam ring bound to a six-membered dihydrothiazine ring. This two-ring system causes distortion of the β-lactam amide bond, resulting in decreased resonance stabilization and increased reactivity. β-lactams inhibit the formation of peptidoglycan cross-links within bacterial cell walls by targeting penicillin-binding proteins or PBPs.
