Tetracyclines

Tetracyclines are a group of broad-spectrum antibiotics, named for the four hydrocarbon rings common in their structure. Tetracyclines are further defined as a subclass of polyketides or derivatives of polycyclic naphthacene carboxamide. Tetracycline antibiotics act as protein synthesis inhibitors, inhibiting the binding of aminoacyl-tRNA to the mRNA-ribosome complex. They do so mainly by binding to the 30S ribosomal subunit in the mRNA translation complex.

Description

Chlortetracycline, isolated from Streptomyces aureofaciens, was the first tetracycline antibiotic discovered. It has shown the most efficacy against gram-positive organisms, but is also effective on a variety of gram-negative bacteria. Resistance to chlortetracycline arises from a loss of cell wall permeability.

Description

Doxycycline hyclate is a semisynthetic tetracycline antibiotic effective against bacteria such as Mycoplasma pneumoniae and Haemophilus influenzae. It is also a broad-spectrum inhibitor of matrix metalloproteases (MMPs). Doxycycline hyclate is routinely used to select for cells containing the pcDNA3.1 resistance plasmid in cell line 293 at an effective concentration of 2 μg/mL. It is freely soluble in aqueous solution.
