Penicillins

Penicillins are a group of antibacterial agents derived from the Penicillium fungus and includes penicillin G, penicillin V, ampicillin and carbenicillin. Penicillins are β-lactams, which interfere with PBP (Penicillin Binding Protein) activity involved in the final phase of peptidoglycan synthesis. PBPs are enzymes that catalyze a pentaglycine cross-link between alanine and lysine residues providing additional strength to the cell wall. Without a pentaglycine cross-link, the integrity of the cell wall is severely compromised and ultimately leads to cell lysis and death. Many penicillins, such as ampicillin, amoxicillin and carbenicillin, are commonly used as selective agents in laboratory, bacterial screening protocols.

6-Aminopenicillanic acid

Catalog ID Size Pricing
A-915-10 10 g $ 54.00
A-915-25 25 g $ 88.00

Description

6-Aminopenicillanic acid (6-APA) is the central component of penicillin β-lactam antibiotics which are generated by Penicillium. 6-APA forms from penicillin G after being catalyzed by penicillin acylases. This compound can be used to create semi-synthetic antibiotics that are effective against non-penicillinase forming bacterium, such as Streptococcus and some Staphylococcus.

6-APA is used as precursor for antibiotic compounds ampicillin and amoxicillin. Antibiotic synthesis of 6-APA can be augmented with a supersaturated solution of nucleophiles. Other methods for enhancing effectiveness involves diminishing ionic strength in a reaction mixture along with the addition of 2-butanol. Implementing a pH gradient starting at pH 7 and reducing the pH to 6.5 near reaction completion can greatly increase reaction output.

Ampicillin/Sulbactam (2:1)

Catalog ID Size Pricing
A-115-1 1 g $ 224.00

Description

Ampicillin/sulbactam (2:1) is a broad-spectrum combination of a β-lactam antibiotic with a β-lactamase inhibitor. It is used for in vitro susceptibility tests in microbiology, pharmacokinetics and pharmacodynamics studies.

Ampicillin binds to the active site of penicillin binding proteins (PBP), preventing peptidoglycan cross-link formation of the bacterial cell wall. Inhibition of PBPs weakens the bacterial cell wall and leads to cell lysis. Sulbactam irreversibly inhibits β-lactamase enzymes generated by bacteria, preserving ampicillin activity and enhancing potency.

Description

Carbenicillin is a member of the penicillin β-lactam antibiotics. However, unlike most β-lactams, carbenicillin disodium is limited to primarily gram-negative bacteria including Pseudomonas aeruginosa and common enteric species. It can be used in selection protocols and is often preferred over ampicillin due to its increased stability. This allows for less contamination of unwanted bacterial colonies. In addition, carbenicillin has been used to regulate bacterial growth in plants, yielding low regeneration frequencies. It can be used in genetic transformation applications to select for AmpR transformed cells.

Cloxacillin Sodium

Catalog ID Size Pricing
C-805-1 1 g $ 58.00
C-805-5 5 g $ 239.00

Description

Cloxacillin sodium is a sodium salt formed from a chlorinated semisynthetic derivative of penicillin. It has been shown to exhibit significant bactericidal activity against penicillin G-resistant staphylococci.

Penicillins are a type of β-lactam antibiotic consisting of a four-membered β-lactam ring bound to a five-membered thiazolidine ring. This two-ring system causes distortion of the β-lactam amide bond, resulting in decreased resonance stabilization and increased reactivity. β-lactams inhibit the formation of peptidoglycan cross-links within bacterial cell walls by targeting penicillin-binding proteins or PBPs. Consequently, the bacterial cell wall becomes weak and cytolysis occurs. Resistance to β-lactam antibiotics occurs in the presence of cells containing plasmid encoded extended spectrum β-lactamases or ESBLs.

Antibiotics are often used in clinical in vitro tests known as antimicrobial susceptibility tests or ASTs to determine their efficacy against certain bacterial species. They are tested against gram-negative and gram-positive bacteria using panels, discs, and MIC strips by medical microbiologists. ASTs decrease the risk of using an antibiotic against bacteria exhibiting resistance to it, and the results are used in clinical settings to determine which antibiotic(s) to prescribe for various infections.
