Description
FGF1 is a member of the Fibroblast Growth Factor family of proteins. Also called FGF-acidic or aFGF, FGF1 lacks signal peptides and is believed to be secreted by a nonclassical mechanism. It has been detected in large amounts in the brain and is expressed in hepatocyte cells, vascular smooth muscle cells, CNS neurons, skeletal muscle cells, fibroblasts, keratinocytes, endothelial cells, intestinal columnar epithelial cells, and pituitary basophils and acidophils. FGF1 exhibits considerable species cross-reactivity and stimulates the proliferation of cells of mesodermal origin, as well as many cells of neuroectodermal, ectodermal, and endodermal origin. It is most closely associated with FGF2 (bFGF) (see additional information).

Description
FGF2 is a member of the fibroblast growth factor family of proteins. It is often called FGF-basic or bFGF. FGF2 is a single-chain polypeptide growth factor that plays a significant role in wound healing, it is a potent inducer of angiogenesis and it is essential for the growth of many stem and progenitor cells. FGF2 shares a 55% amino acid residue identity with FGF1 and interacts with heparin sulfate with nanomolar affinity. It is a potent inducer of DNA synthesis in a variety of cell types. FGF2 activates mesenchymal splice variants of FGF receptors (c splice variants) and also activates FGF Receptor 1b, but has little activity against FGFR2b or FGFR3b. It is most closely associated with FGF1 (aFGF) (see additional information).

Description
FGF10/KGF2 is a member of the Fibroblast Growth Factor family of proteins. KGF2 cDNA encodes a 208 amino acid residue protein with a hydrophobic amino-terminal signal peptide. Human KGF2 shares 92% and 95% amino acid sequence identity with mouse and rat KGF2, respectively. In the FGF family, KGF2 is most closely related to FGF7 (KGF1), FGF3 and FGF22 (see additional information). The expression of KGF2 transcript has been shown to be most abundant in the embryo and adult lung. Recombinant KGF2 has been shown to be mitogenic for epithelial and epidermal cells but not fibroblasts. KGF2 may play unique roles in brain and lung development, wound healing and limb bud formation.

Description
FGF13 is a member of the Fibroblast Growth Factor family of proteins. It is one of four FGFs that act as intracellular signaling molecules, independent of FGF receptors. They have instead been shown to interact with voltage-gated ion channels and the MAPK scaffold protein, islet-brain 2. FGF13 is expressed in the developing and adult central nervous system and acts to stabilize microtubules. It is required for axon and leading process development, as well as neuronal migration. It is believed to be a candidate gene for syndromal and nonspecific forms of X-chromosome-linked intellectual disability (XLID).

Description
FGF18 is a member of the Fibroblast Growth Factor family of proteins. FGF18 has been shown in vitro to induce neurite outgrowth in PC12 cells. Studies of the similar proteins in mouse and chick suggest that this protein is a pleiotropic growth factor that stimulates proliferation in tissues such as the liver and small intestine. FGF18 acts as a trophic factor for elastic chondrocytes in vivo and articular chondrocytes cultured in vitro and plays an autocrine role in normal articular cartilage repair. FGF18 –/– mice have decreased expression of osteogenic markers and delayed long-bone ossification. FGF18 has been shown in vitro that this protein is able to induce neurite outgrowth in PC12 cells. It is most closely related to FGF8 and FGF17 (see additional information).

Description
FGF19 is a member of the Fibroblast Growth Factor family of proteins. It is in the same endocrine signaling subfamily as FGF21 and FGF23 and is the human ortholog of mouse FGF15. FGF19 is a heparin-dependent ligand and binds exclusively to FGFR4. It is essential for brain and heart development, as well as the metabolism of bile acid, hepatic protein, and glycogen. FGF19 mRNA is expressed in several tissues including fetal cartilage, skin, and retina, as well as adult gall bladder and is overexpressed in a colon adenocarcinoma cell line.

FGF21 is a member of the Fibroblast Growth Factor family of proteins. It is in the same endocrine signaling subfamily as FGF19 and FGF23. FGF21 is a heparin-binding ligand for FGFR1c and FGFR4 that requires the presence of beta-klotho. Expression of FGF21 mRNA has been observed in the liver, the thymus, and several types of adipose tissue. It has been shown to stimulate glucose incorporation in differentiated adipocytes, independent of insulin activity. It has been suggested that FGF21 also acts as key regulator in the adaptation to stress.

FGF23 is a member of the Fibroblast Growth Factor family of proteins. In the FGF family, FGF23 is most closely associated with FGF15/19 and FGF21 (see additional information). FGF23 shares 71% homology with mouse Fgf23 at the amino acid level. FGF23 is produced by osteocytes and osteoblasts in response to high circulating phosphate levels, elevated parathyroid hormone, and circulatory volume loading. It functions as an endocrine phosphatonin by suppressing circulating phosphate levels. FGF23 interaction with renal proximal tubular epithelium decreases the renal resorption of phosphate by down regulating phosphate transporters and by suppressing vitamin D production. It also decreases the intestinal absorption of phosphate.

FGF4 is a member of the Fibroblast Growth Factor family of proteins and was first identified by its oncogenic activity. It plays an important role in embryonic development, acting as an autocrine or paracrine ligand through the FGFR 1c, 2c, 3c, and 4 receptors to support stem cell self-renewal. It is believed that FGF4, with the combined activity one or more apical ectodermal ridge FGFs (FGF2, FGF4, FGF8, and FGF9) works in a positive feedback loop with SHH to control limb development.
