Description

FAM19A2 (TAFA-2) is chemokine-like protein containing ten cysteine residues that are conserved amongst the FAM19A (TAFA) family. This family of proteins was first identified by using a clustering algorithm on uncharacterized cDNA sequences and found to be distantly related to C-C motif chemokines, such as CCL3. FAM19A2 is expressed predominantly in the CNS, with very low expression levels detected in colon and spleen. Expression levels of FAM19A2 are highest in the occipital and frontal cortex and it is believed to be expressed constitutively. FAM19A2 is hypothesized to function as a brain-specific chemokine that regulates immune response in the CNS, as a neurokine that modulates immune response, or as a control of axonal sprouting following brain injury.

FGF13 is a member of the Fibroblast Growth Factor family of proteins. It is one of four FGFs that act as intracellular signaling molecules, independent of FGF receptors. They have instead been shown to interact with voltage-gated ion channels and the MAPK scaffold protein, islet-brain 2. FGF13 is expressed in the developing and adult central nervous system and acts to stabilize microtubules. It is required for axon and leading process development, as well as neuronal migration. It is believed to be a candidate gene for syndromal and nonspecific forms of X-chromosome-linked intellectual disability (XLID).

FGF18 is a member of the Fibroblast Growth Factor family of proteins. FGF18 has been shown in vitro to induce neurite outgrowth in PC12 cells. Studies of the similar proteins in mouse and chick suggest that this protein is a pleiotropic growth factor that stimulates proliferation in tissues such as the liver and small intestine. FGF18 acts as a trophic factor for elastic chondrocytes in vivo and articular chondrocytes cultured in vitro and plays an autocrine role in normal articular cartilage repair. FGF18 –/– mice have decreased expression of osteogenic markers and delayed long-bone ossification. FGF18 has been shown in vitro that this protein is able to induce neurite outgrowth in PC12 cells. It is most closely related to FGF8 and FGF17 (see additional information).

Description

FGF23 is a member of the Fibroblast Growth Factor family of proteins. In the FGF family, FGF23 is most closely associated with FGF15/19 and FGF21 (see additional information). FGF23 shares 71% homology with mouse Fgf23 at the amino acid level. FGF23 is produced by osteocytes and osteoblasts in response to high circulating phosphate levels, elevated parathyroid hormone, and circulatory volume loading. It functions as an endocrine phosphatonin by suppressing circulating phosphate levels. FGF23 interaction with renal proximal tubular epithelium decreases the renal resorption of phosphate by down regulating phosphate transporters and by suppressing vitamin D production. It also decreases the intestinal absorption of phosphate.
